STAT3 directly binds to miRNA promoters to upregulate their expression, forming STAT3/miR-223/IL-6 or STAT3/miR-29a-5p loops. In CSSC and CAC, STAT3 upregulates miRNA expression by binding to the miR-223 and miR-29a-5p promoters. Subsequently, exosomal miR-223 induces monocytes/macrophages to secrete IL-6, which in turn mediates the upregulation of STAT3 expression in CSSC cells. Meanwhile, overexpression of miR-29a-5p also increases the expression of STAT3. Finally, a positive feedback loop is formed to promote the growth of CSCC and CAC tumors
