CAA acquisition method. Adipocytes were obtained from 3T3L1 mouse preadipocytes by differentiation based on cell treatment with a mixture of rosiglitazone, insulin, dexamethasone, and 3-isobutyl-1-methylxanthine (IBMX) for 48 h, followed by incubation with insulin for another 48 h. Then, adipocytes were cultured for 7 days in the presence of melanoma cells seeded on Transwell inserts in a combination of DMEM with 10% BCS and DMEM with 10% FBS (1:1 ratio). The 3T3L1 fibroblasts and control adipocytes were also grown in such a medium. Next, cells were harvested and used for further experiments, or the medium was changed for DMEM without serum, collected after 72 h, and then used for analysis of extracellular proteins level.
